Hi
You need to add a reference to that toolkit dll in your application.
or
In the design view of any aspx page, try to right click on the toolbox general section or any section then select choose items
browse to select that dll you have downloaded. Then press ok. You will see all the toolkit controls in your toolbox. Now if you drag and drop it will add the reference for you.

Thanks,
Debata

You can try this
Copy that ajax control toolkit dll to bin folder or add a reference in case you are using web application project.
Then after page directives
<%@ Register Assembly="AjaxControlToolkit" Namespace="AjaxControlToolkit" TagPrefix="act" %>

Now if will write in <act: aspx page you will see ajax control tool kit
