There is a well written article on this website http://www.dotnetfunda.com/articles/article870-a-walkthrough--to-aspnet-40-url-routing-.aspx, after going through this, you should be able to understand (this is based on ASP.NET 4.0 and recommended to use it).

Or also read http://www.dotnetfunda.com/articles/article75.aspx (older approach however works in the ASP.NET 2.0 and 3.5).
