authorization programatically

I posted this to this site but my post seemed to be gone.
I can't find a solution yet for this.
I'd like to use 'authorization' in location programatically on the fly. I saw a sample which shows how to do it. It doesn't use updatding the contents of web.config etc.
How can it be done?
